ANTIBACTERIAL AGENT.

HYPOTHESIS
METHODOLOGY
RESULTS
Mangifera indica is a member of the family Anacardiaceae.(Ian S.E.
Bally, 2006). The major benefit of the seed kernel is where Kernel fat
from the seed is used to cure stomatitis. Kernel powder is used as a
vermifuge, for hemorrhages, bleeding hemorrhoids, bacterial agent and
diarrhea. (Ian S.E. Bally, 2006). Two tea spoon full of kernel of mango
seed is cooked with one glass of water and the mixture is made into a
semisolid paste form. (Dr. Janardhana V. Hebbar). Previous research
focuses exclusively on the antibacterial activity of mango kernel ex-
tracts against several Gram-positive and Gram-negative bacterial
strains. Method applied in this research was the agar well diffusion
method. The antibacterial properties of mango seed kernel extracts are
investigated by using the agar well diffusion method, the Minimum In-
hibitory concentration (MIC) of mango seed kernels. The outcome of
this research was successful with prominent significance.(M.E.S. Mir-
ghani. et al, 2009). Therefore this study is done to confirm the signifi-
cance of mango seed kernel as an antibacterial agent.
General objectives:
To investigate any antibacterial activity of the extraction
from the Mangifera indica seed kernel.
Specific Objectives:
i) To compare the antibacterial properties of Mangifera indica seed
kernel extract.
ii) To compare the antibacterial activity of mango kernel extract
against Staphylococcus aureus and Escherichia coli.
Iii) To investigate the antibacterial activity using the disk diffusion
method.
Mangifera indica seed kernel extract would probably become an al-
ternative source of new and neutral antibacterial agent.
Mango fruit pur-
chased and seed
dried. Than, kernel
taken out and grind.
Solvents prepared
are 85% acetone and
85% methanol.
1gram of sample is placed
into a tube and 1ml and
1.5ml solvents respectively
are pipette into the tube.
Centrifuge at
13,000rpm for 15 min-
utes, supernatant trans-
ferred into new tube.
Bacteria inoculated onto
agar plate. Plain disk
containing extract is
placed onto the plate.
Incubate plate over-
night, “zone of inhibi-
tion” is observed. Pro-
ceed with MIC.
From the observation and the measurement of the diameter of inhibi-
tion zone formed, it has been demonstrated that the extracts from
Mangifera indica seed kernel has significance and the potential of acting
as an antibacterial agent. The results shows that The extract works well
against Staphylococcus aureus and Escherichia coli. Antibacterial activity
against Gram-positive bacterial strain Staphylococcus aureus gave
higher results compared to Gram-negative bacterial strain Escherichia
coli. This may be because of the compositional values of the seed kernel,
further studies may be required to confirm the effectiveness. Comparison
to the positive control, which is the known antibiotic tetracycline, the di-
ameter of inhibition was slightly larger and very significant. P-Value
< 0.05. The significance of tetracycline is better because Tetracycline is
already recognized as an wide spectrum antibiotic which inhibits the
growth of both Gram-positive bacterial strain Staphylococcus aureus and
Gram-negative bacterial strain Escherichia coli. Minimum Inhibition Con-
centration (MIC) results showes that as the extract concentration de-
creases, the size of the inhibition zone also decreases.
The extracts have proven to act as an antibacterial agent.
I would like to thank my sole supervisor Miss Norshafarina Shari @
Kamaruddin for the guidance, advice, encouragement and endless sup-
port. Her patients is highly appreciated. The knowledge that she has
passed to me is priceless and I will cherish it all my life.
