ā€¢ Filtration: It may be deļ¬ne as a process of separation of
solids from a ļ¬‚uid by passing the same through a porous
medium that retains the solids but allows the ļ¬‚uid to pass
through.

ā€¢ The ļ¬‚uid that passes through is called a ļ¬ltrate.

ā€¢ Filtration is the best established and most versatile method
for removing insoluble from dilute streams like
fermentation broths.
FILTERATION
The separation of solids from a suspension in a liquid by
means of a porous medium or screen which retains the
solids and allows the liquid to pass is termed filtration.

Principle of Filtration
Since the filter medium is permeable only to the fluid, it retains the
solid particles and permits only the fluid to pass through which is
collected as the filtrate. The volume of filtrate collected per unit time
(dV/dt) is termed as the rate of filtration.
As the filtration proceeds, solid particle accumulate on the filter
medium forming a packed bed of solids, called filter cake.
As the thickness of the cake increases
! resistance to flow of filtrate increases
! rate of filtration gradually decreases.
If rate is maintained to be constant then
pressure difference driving force (-Ī”P) will
increase.
Therefore, a batch filter is operated either at constant pressure or at constant rate.
Theories of ļ¬ltration
ā€¢ The ļ¬‚ow of liquid through a ļ¬lter follows the basic rules that govern the
ļ¬‚ow of any liquid through the medium oļ¬€ering resistance.

ā€¢ The rate of ļ¬‚ow may be expressed as-

ā€¢ Rate = driving force / resistance

ā€¢ The rate of ļ¬ltration may be expressed as volume (litres) per unit time
(dv/dt).

ā€¢ Driving force = pressure upstream ā€“ pressure downstream

ā€¢ Resistance is not constant.

ā€¢ It increases with an increase in the deposition of solids on the ļ¬lter
medium.

ā€¢ Therefore ļ¬ltration is not a steady state.

ā–  The rate of flow will be greatest at the beginning of filtration
process, since the resistance is minimum.
ā–  After forming of filter cake, its surface acts as filter medium and
solids continuously deposit adding to thickness of the cake.

Poiseullieā€™s Equation
ā€¢ Poiseullie considered that ļ¬ltration is similar to the streamline

ā€¢ ļ¬‚ow of liquid under pressure through capillaries.

ā€¢ Poiseullieā€™s Equation is-

ā€¢ Where, V = rate of ļ¬‚ow, m3/s 

ā€¢ Ī”P= Pressure diļ¬€erence across the ļ¬lter, Pa 

ā€¢ r	 = radius of capillary in the ļ¬lter bed, m

ā€¢ L = thickness of ļ¬lter cake (capillary length), m

ā€¢ š¶ = viscosity of ļ¬ltrate

ā€¢ If the cake is composed of bulky mass of particles and the liquid ļ¬‚ows
through the interstice, then ļ¬‚ow of liquids through these may be expressed by
this equation.
Darcyā€™s Equation
ā€¢ Poiseullie's law assumes that the capillaries found in the ļ¬lter are highly irregular
and non-uniform.

ā€¢ Therefore, if the length of capillary is taken as the thickness of bed, a correction
factor for radius is applied so that the rate is closely approximated and
simpliļ¬ed.

ā€¢ The factors inļ¬‚uencing the rate of ļ¬ltration has been incorporated into an
equation by Darcy, which is:

ā€¢ Where, K = permeability coeļ¬ƒcient of cake, m2

ā€¢ A	= surface area of porous bed (ļ¬lter medium), m2 

ā€¢ Other terms are same as previous equation

ā€¢ K depends on characteristics of cake, such as porosity, speciļ¬c

ā€¢ surface area and compressibility.

Filter Media
ā€¢ The surface upon which solids are deposited in a ļ¬lter is called the
ā€œFilter mediumā€

ā€¢ Properties of ideal ļ¬lter medium:

ā€¢ It should-

ā€¢ be capable of delivering a clear ļ¬ltrate at a suitable production rate.

ā€¢ have suļ¬ƒcient mechanical strength.

ā€¢ be inert.

ā€¢ retain the solids without plugging at the start of ļ¬ltration.

ā€¢ Not absorb dissolve material.

ā€¢ Sterile ļ¬ltration imposes a special requirement since the pore size
must not exceed the dimension of bacteria or spores.
Material used as ļ¬lter media
Woven material

ā€¢ Made up of wool, silk, metal or synthetic ļ¬bres (rayon, nylon
etc.).

ā€¢ These include a) wire screening and b) fabrics of cotton,
wool, nylon.

ā€¢ Wire screening e.g. stainless steel is durable, resistance to
plugging and easily cleaned.

ā€¢ Cotton is a common ļ¬lter ,however, Nylon is superior for
pharmaceutical use, since it is unaļ¬€ected by mold, fungus
or bacteria and has negligible absorption properties .

ā€¢ The choice of ļ¬bre depends on chemical

ā€¢ Stainless steel plates have pores which act as channels as
in case of meta ļ¬lters.

Bed of granular solid

ā€¢ In some processes, a bed of graded solids may be formed
to reduce resistance of ļ¬‚ow.

ā€¢ Ex. Of granular solids are gravel, sand, asbestos, paper
pulp and keiselgur.

ā€¢ Choice of solids depends on size of solids in process.
Membrane ļ¬lter media

ā€¢ These are cartridge units and are economical and available in pore size
of 100 Ī¼m to even less than 0.2 Ī¼m.

ā€¢ Can be either surface cartridges or depth type cartridges.

Surface cartridges:
ā€¢ These are corrugated and resin treated papers and used in hydraulic
lines.

ā€¢ Ceramic cartridges and porcelain ļ¬lter candles are examples.

ā€¢ Can be reuse after cleaning.

Depth type cartridges:
ā€¢ Made up of cotton, asbestos or cellulose.

ā€¢ These are disposable items, since cleaning is not feasible.
Prefabricated porous solid units

ā€¢ Used for its convenience and eļ¬€ectiveness.

ā€¢ Sintered glass, sintered metal, earthenware and porous
plastics are used for fabrication.
Filter aids
ā€¢ The objective of ļ¬lter aid is to prevent the medium from
becoming blocked and to form an open, porous cake, hence,
reducing the resistance to ļ¬‚ow of the ļ¬ltrate.

ā€¢ Filter aid forms a surface deposit which screens out the solids
and also prevents the plugging of supporting ļ¬lter medium.

ā€¢ Characteristics of ļ¬lter aids:

ā€¢ Chemically inert and free from impurities.

ā€¢ Low speciļ¬c gravity, so remain suspended in liquids.

ā€¢ Porous rather than dense, so that pervious cake can be
formed.

ā€¢ Recoverable.
Disadvantages:

ā€¢ Remove the coloured substances by absorbing them.

ā€¢ Sometimes active principles such as alkaloids are
absorbed on ļ¬lter aid.

ā€¢ Rarely, ļ¬lters are source of contamination such as
soluble iron salts,
Handling of ļ¬lter aids
ā€¢ Filter aids may be used in either or both two ways:

ā€¢ Pre- coating technique: by forming a pre-coat over the
ļ¬lter

ā€¢ medium by ļ¬ltering a suspension of the ļ¬lter aid .

ā€¢ Body- mix technique: A small proportion of the ļ¬lter aid
(0.1-

ā€¢ 0.5 %) is added to the slurry to be ļ¬ltered. This slurry is
recirculated through the ļ¬lter until a clear ļ¬ltrate is
obtained, ļ¬ltration then proceeds to completion.
ā€¢ Diļ¬€erent ļ¬‚ow rates can be achieved depending on grade of aid-

ā€¢ Low ļ¬‚ow rate: ļ¬ne grade ļ¬lter aids- mainly used for clarity

ā€¢ Fast ļ¬‚ow rate: coarse grade ļ¬lter aids- acceptable ļ¬ltrate.

Examples of ļ¬lter aids

ā€¢ Diatomite (Keiselgur) , obtained from natural siliceous deposits.

ā€¢ Perlite, Cellulose, charcoal, talc, bentonite , fullers earth etc.

Principle:

ā€¢ Mechanism is surface ļ¬ltration.

ā€¢ The slurry enters the frame by pressure and ļ¬‚ows
through ļ¬lter medium.

ā€¢ The ļ¬ltrate is collected on the plates and send to outlet.

ā€¢ A number of frames and plates are used so that surface
area increases and consequently large volumes of slurry
can be processed simultaneously with or without
washing.

Construction
ā€¢ The Filter press is made of two types of units, plate and
frames.

ā€¢ Usually made of aluminium alloy.

ā€¢ Sometimes,	 these	 are	also	 lacquered	 for	protection	
against corrosive chemicals and made suitable for steam
sterilization.

Frame

ā€¢ It contains a open space inside wherein the slurry reservoir
is maintained for ļ¬ltration and an inlet to receive the slurry.

ā€¢ It is indicated by two dots in description.

ā€¢ It is selected based on the thickness of cake formed
during ļ¬ltration.

ā€¢ Optimum thickness of frame should be chosen.

Plate

ā€¢ The plate has a studded or grooved 

surface to support the ļ¬lter cloth 

and an outlet.

ā€¢ It is indicated by one dot in description.

ā€¢ Plate supports the ļ¬lter medium, receiving the ļ¬ltrate and outlet.

ā€¢ The ļ¬lter medium usually cloth is interposed between plate and
frame.

ā€¢ Plate, ļ¬lter medium, frame are arranged in sequence and clamed to
a supporting structure.

ā€¢ It is normally described by dots as 1.2.1.2.1 so on.

ā€¢ A number of plates and frames are employed so that the ļ¬ltration
area is as large as necessary.

ā€¢ Number of ļ¬ltration units are operated in parallel.

ā€¢ Channels for slurry inlet and ļ¬ltrate outlet can be arranged by ļ¬tting
eyes to the plates and frames, these join together to form a channel.

ā€¢ In some types only one inlet channel is formed, while each plate is
having individual outlets controlled by valves.

Washing operation
ā€¢ When washing of cake is also required modiļ¬ed plate and
frame ļ¬lter is used.

ā€¢ For this purpose an additional channel is included called as
washing plate and are identiļ¬ed by 3 dots.

ā€¢ In the half of the washing plate, there is a connection from
wash water cannel to the surface of plate.

ā€¢ The sequence of arrangement of plates and frames can be
represented by dots as 1.2.3.2.1.2.3.2.1 so on (between 1
and 1, 2.3.2 must be arranged.
Filter leaf
ā€¢ Principle:

ā€¢ It is an apparatus consisting of a longitudinal drainage
screen covered with a ļ¬lter cloth.

ā€¢ The mechanism is surface ļ¬ltration and acts as sieve or
strainer.

ā€¢ Vacuum	 or	 pressure	can	 be	applied	 to	 increase	
the	rate	 of ļ¬ltration.
ā€¢ Construction:

ā€¢ The	leaf	 ļ¬lter	 is	 consisting	 of	 a	 frame	enclosing	
a	 drainage screen or grooved plate.

ā€¢ The frame may be any shape circular, square or
rectangular.

ā€¢ The whole unite being covered with ļ¬lter cloth.

ā€¢ The outlet for	 the ļ¬ltrate connects	 to the	inside	of the
frame through suction.

Advantages
ā€¢ Simplest form of ļ¬lter used for batch process.

ā€¢ A number of units can be connected in parallel to increase
the surface area of ļ¬ltration.

ā€¢ Pressure diļ¬€erence can be obtained either with vacuum or
using pressure up to the order of 800 kilopascals.

ā€¢ Labour costs for operating the ļ¬lter leaf are fairly moderate.

ā€¢ The eļ¬ƒciency of washing is high.

ā€¢ The slurry can be ļ¬ltered from any vessel.

ā€¢ The cake can be washed simply by immersing the ļ¬lter in a
vessel of Water.
Meta ļ¬lter
Principle:

ā€¢ Mechanism is surface ļ¬ltration.

ā€¢ In this, metal rings contain semicircular projections,
which are arranged as a nest to form channels on the
edges.

ā€¢ This	 channel	 oļ¬€ers	resistance	 (strainer)	to	 the	
ļ¬‚ow	 of	 solids (coarse particles).

ā€¢ The clear liquid is collected into receiver from the top.
Construction
ā€¢ Metaļ¬lter consists of a series of metal rings.

ā€¢ These are threaded so that a channel is formed on the edges.

ā€¢ It contains a grooved drainage column on which a series of metal
rings are packed.

ā€¢ These rings are usually made up of stainless steel and have
dimensions of about 15.0 mm internal diameter and 22.0 mm external
diameter.

ā€¢ Each metal ring has a number of semicircular projections (0.8 mm in
thickness) on one side of surface.

ā€¢ The projections are arranged as a nest to form channels on the edges.

ā€¢ These rings are tightened on the drainage column with a nut.

ā€¢ Metaļ¬lters are also known as edge ļ¬lters.

ā€¢ Clariļ¬cation of insulin liquors

Advantages 

ā€¢ Can be used under high pressures, without any danger of bursting the
ļ¬lter medium.

ā€¢ Running cost are low, as separate ļ¬lter medium is not used.

ā€¢ Can be constructed from a material that can provide excellent
resistance to corrosion and avoid contamination of sensitive products.

ā€¢ It is extremely versatile ļ¬lter because ļ¬ne as well as large both type of
particles can be separated.

ā€¢ Removal of cake can be carried out by simply back- ļ¬‚ushing with
water.

ā€¢ Change over from one batch to another or one product to another is
easy.

ā€¢ Sterile products can be handled.
Cartridge ļ¬lter
Principle:

ā€¢ It is a thin porous membrane in which pre ļ¬lter and
membrane ļ¬lter are combined in a single unit.

ā€¢ The ļ¬ltration action is mainly sieve like and particles
are retained on the surface.
Construction

ā€¢ It has cylindrical conļ¬guration made with disposable or
changeable ļ¬lter media.

ā€¢ Made up of either plastic or metal.

ā€¢ Consist of two membrane ļ¬lters (sieve like) made of
polypropylene: pre-ļ¬lter and actual ļ¬lter for ļ¬ltration.

ā€¢ A protective layer surrounds them.

ā€¢ The cartridge are housed in a holder and a number of
cartridges can be placed in a same housing.

ā€¢ The housing is closed with the lid.

ā€¢ Housing has provisions for slurry inlet and outlets.

ā–  Particularly useful for preparation of particulate free
solutions for parenterals and ophthalmic uses.
ā–  This filter holder will process 1000 ā€“
15000 litres of sterile solution per hour.

Advantages:

ā€¢ Autoclaving can be done for sterile operations due to
stainless steel construction.

ā€¢ Cartridge with self cleaning devices are advantageous.

ā€¢ Rapid	disassembling	as	well	 as	reusing	 of	 ļ¬lter	
medium	 is possible.

ā€¢ Cartridge are not brittle, when they are dry.

ā€¢ Used as in-line continuous ļ¬ltration, which reduces
handling of solutions. It minimize chances of
contaminations.
Disadvantages:

ā€¢ A number of manufactures provide the components,
which are generally not interchangeable between
suppliers.

ā€¢ Cost of disposable elements oļ¬€sets the labour saving
in terms of assembly and cleaning of cartridge
clariļ¬ers.
Rotary drum ļ¬lter
Principle:

ā€¢ Slurry	 ļ¬ltered	 through	 sieve	like	 mechanism	on	
the	rotation drum surface, under the condition of
vacuum.

ā€¢ In addition compression, drying (using hot air), and
removing the ļ¬lter cake (using knife) are possible.
Construction:

ā€¢ It consist of a metal cylinder mounted horizontally.

ā€¢ The drum may be up to 3 meters in diameter and 3.5
meters in length and gives surface area of 20 meter
square.

ā€¢ The curved surface being a perforated plate,
supporting a ļ¬lter cloth.

ā€¢ Internally, it is divided into several sectors and a
separate connection is made between each sector
and a special rotary valve.
ā€¢ The drum is dipped into the slurry and vacuum applied to the

ā€¢ outlet, which is connected to the ļ¬ltrate receiver.

ā€¢ When the cake has formed, the cake drained or partially
dried by vacuum.

ā€¢ The drum is sprayed with water to wash the cake.

ā€¢ Retaining the vacuum connection drains the cake and
produces partial dryness then, removed by a doctor knife.

ā€¢ When the solids of the slurry are too much that the ļ¬lter cloth
becomes blocked with the particles, a pre-coat ļ¬lter may be
used.

ā€¢ 	 A pre-coat of ļ¬lter aid is deposited on the drum prior to the
ļ¬ltration process.
USE

ā€¢ The rotary ļ¬lter for continuous operation on large quantities of
slurry.

ā€¢ Suitable for slurry contains considerable amounts of solids in
the range 15-30%.

ā€¢ Examples of pharmaceutical application include

ā€¢ the collection of calcium carbonate, magnesium carbonate,
and starch.

ā€¢ The separation of the mycelium from the fermentation liquor in

ā€¢ the manufacture of antibiotics.
Advantages 

ā€¢ The rotary ļ¬lter is automatic and is continuous in
operation, so that the labour costs are very low.

ā€¢ The ļ¬lter has a large capacity , so it is suitable for the
ļ¬ltration of highly concentrated solutions.

ā€¢ Variation of the speed of rotation enables the cake
thickness to be controlled.

ā€¢ Pre-coat of ļ¬lter aid could used to accelerate the
ļ¬ltration rate.

ā€¢ Filter has large surface area.
Disadvantages 

ā€¢ The rotary ļ¬lter is a complex piece of equipment , with many
moving parts and is very expensive,.

ā€¢ In addition to the ļ¬lter itself, some accessories are connected

ā€¢ ,e.g., a	vacuum	 pump,	vacuum receivers,	 slurry	 pumps	
and agitators are required .

ā€¢ The cake tends to crack due to the air drawn through by the

ā€¢ vacuum system, so that washing and drying are not eļ¬ƒcient.

ā€¢ Being a vacuum ļ¬lter, the pressure diļ¬€erence is limited to 1
bar and hot ļ¬ltrates may boil.

ā€¢ It is suitable only for straight- forward slurries

Deļ¬nition ā€¢ Filtration: It may be deļ¬ne as a process of separation of solids from a ļ¬‚uid by passing the same through a porous medium that retains the solids but allows the ļ¬‚uid to pass through. ā€¢ The ļ¬‚uid that passes through is called a ļ¬ltrate. ā€¢ Filtration is the best established and most versatile method for removing insoluble from dilute streams like fermentation broths.
  • 3. FILTERATION The separation of solids from a suspension in a liquid by means of a porous medium or screen which retains the solids and allows the liquid to pass is termed filtration.
  • 4. Terms used in filtration cake Slurry ā€¢ Suspension to be filtered Filter ā€¢ Porous medium medium used to retain solid Filterā€¢ Accumulated solids on the filter Filtrate ā€¢ Clear liquid passing through the filter
  • 5. Application ā€¢ Production of sterile products: ā€¢ HEPA ļ¬lters or laminar air bench ā€¢ Membrane ļ¬lters. ā€¢ Production of bulk drugs ā€¢ Production of liquid dosage ā€¢ Eļ¬„uents and waste water treatment
  • 6. Principle of Filtration Since the filter medium is permeable only to the fluid, it retains the solid particles and permits only the fluid to pass through which is collected as the filtrate. The volume of filtrate collected per unit time (dV/dt) is termed as the rate of filtration. As the filtration proceeds, solid particle accumulate on the filter medium forming a packed bed of solids, called filter cake. As the thickness of the cake increases ! resistance to flow of filtrate increases ! rate of filtration gradually decreases. If rate is maintained to be constant then pressure difference driving force (-Ī”P) will increase. Therefore, a batch filter is operated either at constant pressure or at constant rate.

Theories of ļ¬ltration ā€¢ The ļ¬‚ow of liquid through a ļ¬lter follows the basic rules that govern the ļ¬‚ow of any liquid through the medium oļ¬€ering resistance. ā€¢ The rate of ļ¬‚ow may be expressed as- ā€¢ Rate = driving force / resistance ā€¢ The rate of ļ¬ltration may be expressed as volume (litres) per unit time (dv/dt). ā€¢ Driving force = pressure upstream ā€“ pressure downstream ā€¢ Resistance is not constant. ā€¢ It increases with an increase in the deposition of solids on the ļ¬lter medium. ā€¢ Therefore ļ¬ltration is not a steady state.

Upstream pressure, P1 ā–  The rate of flow will be greatest at the beginning of filtration process, since the resistance is minimum. ā–  After forming of filter cake, its surface acts as filter medium and solids continuously deposit adding to thickness of the cake. Poiseullieā€™s Equation ā€¢ Poiseullie considered that ļ¬ltration is similar to the streamline ā€¢ ļ¬‚ow of liquid under pressure through capillaries. ā€¢ Poiseullieā€™s Equation is- ā€¢ Where, V = rate of ļ¬‚ow, m3/s ā€¢ Ī”P= Pressure diļ¬€erence across the ļ¬lter, Pa ā€¢ r = radius of capillary in the ļ¬lter bed, m ā€¢ L = thickness of ļ¬lter cake (capillary length), m ā€¢ š¶ = viscosity of ļ¬ltrate ā€¢ If the cake is composed of bulky mass of particles and the liquid ļ¬‚ows through the interstice, then ļ¬‚ow of liquids through these may be expressed by this equation.
