Biofilm samples were collected from water meters and pipes in a drinking water distribution system in southern Sweden. Over 600,000 DNA sequences were obtained and classified. The bacterial communities differed between water meters from households with and without complaints about water quality. Water meters from complaining households had fewer Proteobacteria and more Nitrospira and Pedomicrobium. Biofilm communities also differed between water meters and pipes, with pipes containing more Mycobacterium, Nocardia, Desulfovibrio, and Sulfuricurvum. The European Commission is adopting a Communication to encourage EU member states to review national regulations restricting access to professions. A revised Professional Qualifications Directive will introduce an obligation for member states to mutually evaluate one another's professional regulations. The evaluation will assess whether regulations are justified and proportional or unnecessarily limit mobility, employment and competitiveness. It will be conducted in two clusters covering various professional fields over two years. The Commission expects greater transparency of professional regulations and measures by member states to reduce unjustified barriers to accessing certain professions.

Mental health has been in the news quite a bit lately. Dozens of U.S. states are currently suing Meta for contributing to the youth mental health crisis by inserting addictive features into their products, while the U.S. Surgeon General is touring the nation to bring awareness to the growing epidemic of loneliness and isolation. The country has endured periods of low national morale, such as in the 1970s when high inflation and the energy crisis worsened public sentiment following the Vietnam War. The current mood, however, feels different. Gallup recently reported that national mental health is at an all-time low, with few bright spots to lift spirits.
To better understand how Americans are feeling and their attitudes towards mental health in general, ThinkNow conducted a nationally representative quantitative survey of 1,500 respondents and found some interesting differences among ethnic, age and gender groups.
Technology
For example, 52% agree that technology and social media have a negative impact on mental health, but when broken out by race, 61% of Whites felt technology had a negative effect, and only 48% of Hispanics thought it did.
While technology has helped us keep in touch with friends and family in faraway places, it appears to have degraded our ability to connect in person. Staying connected online is a double-edged sword since the same news feed that brings us pictures of the grandkids and fluffy kittens also feeds us news about the wars in Israel and Ukraine, the dysfunction in Washington, the latest mass shooting and the climate crisis.
Hispanics may have a built-in defense against the isolation technology breeds, owing to their large, multigenerational households, strong social support systems, and tendency to use social media to stay connected with relatives abroad.
Age and Gender
When asked how individuals rate their mental health, men rate it higher than women by 11 percentage points, and Baby Boomers rank it highest at 83%, saying it’s good or excellent vs. 57% of Gen Z saying the same.
Gen Z spends the most amount of time on social media, so the notion that social media negatively affects mental health appears to be correlated. Unfortunately, Gen Z is also the generation that’s least comfortable discussing mental health concerns with healthcare professionals. Only 40% of them state they’re comfortable discussing their issues with a professional compared to 60% of Millennials and 65% of Boomers.
Race Affects Attitudes
As seen in previous research conducted by ThinkNow, Asian Americans lag other groups when it comes to awareness of mental health issues. Twenty-four percent of Asian Americans believe that having a mental health issue is a sign of weakness compared to the 16% average for all groups. Asians are also considerably less likely to be aware of mental health services in their communities (42% vs. 55%) and most likely to seek out information on social media (51% vs. 35%).

Der Citratzyklus
Der Citratzyklus wird nach seinemEntdecker Dr. Krebs entsprechend auch Krebszyklus genannt. In
ihm laufen die Abbauwege aller energieliefernden Stoffe (Kohlenhydrate, Fette, Proteine)
zusammen um auf die gemeinsame Endstrecke, die Atmungskette überführt zu werden.
Gleichzeitig dient der Citratzyklus von seinen Zwischenprodukten ausgehend auch dem Aufbau
vieler wichtiger Moleküle (z.B. Aminosäuren,
Nucleotidbasen, Cholesterin usw.)
Er kann also nicht nur katabolisch (abbauend) sondern auch anabolisch (aufbauend) wirken.
Solche Stoffwechselwege nennt man dann amphibol
( amphi: gr. Vorsilbe für beid- , doppel-).
Nicht umsonst wird er auch als der Mittelpunkt des Zellstoffwechsels bezeichnet.
Lokalisiert ist der Citratzyklus in den Mitochondrien. Hier finden u. a. auch der Fettsäuren-Abbau
und die Atmungskette statt. Diese lokale Verbindung verdeutlicht das Zusammenspiel der drei
Stoffwechselprozesse untereinander.
2. Citratzyklus 20.12.2015 EuSaB e.V.
1. Acetyl-CoA – die Ausgangssubstanzdes Citratzyklus
Acetyl steht für die Rest-Gruppe der Essigsäure, diese wird dem Citratzyklus gebunden an das
Coenzym A geliefert. Man spricht dann von aktivierter Essigsäure, da die entstandene
Thioestherbindung hoch energetisch ist und somit ein hohes Übertragungspotential für die
AcetylGruppe aufweist.
Acetyl-Reste werden zu Kohlendioxid, unserem Stoffwechselendprodukt, oxidiert. Kohlendioxid ist
in unserem Organismus die Endstation, da das Kohlenstoff-Atom an zwei Sauerstoff-Atome
gebunden ist und somit seine höchste Oxidationsstufe erreicht hat. Das bedeutet für unseren
Organismus, dass aus dieser Verbindung keine Energie mehr rauszuholen ist.
Aufgrund des pH-Wertes von 7,4 liegt die Essigsäure in unserem Körper meist dissoziiert vor, d. h.
sie gibt ein Proton in die Lösung ab. Man spricht dann vom Acetat der Essigsäure.
Der Grossteil der Acetyl-Reste stammt aus der b-Oxidation der Fettsäuren und der Pyruvat-
Dehydrogenase-Reaktion.
Aminosäuren werden teilweise direkt zu Acetyl-CoA abgebaut, manche gehen erst noch den Weg
über das Pyruvat oder steigen mit ihren Abbauprodukten erst als „Zwischenstationen“ des
Citratzyklus ein.
Daraus lässt sich erkennen das früher oder später alle Substanzen der drei großen Klassen von
Brennstoffmolekülen (Kohlenhydrate, Aminosäuren, Fettsäuren) die unser Körper aufnimmt im
Citratzyklus landen, wenn sie aerob abgebaut werden sollen.
1.1 Die b-Oxidation der Fettsäuren
Beim Abbau von Fettsäuren (langkettige Kohlenwasserstoffverbindungen mit einer Carboxy-
Gruppe am a-C-Atom) werden immer die zwei ersten C-Atome mit dem Coenzym A versehen und
„abgeknipst“. Das b im Namen rührt einfach daher, das bei einer der aus vier Teilen bestehenden
Reaktion das b-C-Atom oxidiert wird. Es entsteht bei der b-Oxidation Acetyl-CoA , welches für den
Citratzyklus genutzt werden kann.
1.2 Die Pyruvat-Dehydrogenase-Reaktion
Pyruvat (= Brenztraubensäure) ist das Endprodukt der Glykolyse und entsteht auch beim Abbau
einiger Aminosäuren. Durch einen großen
Multienzymkomplex, der Pyruvat-Dehydrogenase (PDH) wird aus dem Pyruvat und dem Coenzym
A ® Acetyl-CoA. Dabei wird Kohlendioxid abgespalten und Energie in Form von einem Molekül
NADH/H+ gespeichert.

Acetyl-CoA wird nur in den Mitochondrien gebildet und kann diese eigenständig auch nicht
verlassen. Dazu später mehr.
Wichtig: der Schritt von Pyruvat zu Acetyl-CoA ist irreversibel für unseren Organismus. Dies hat
zur Konsequenz, dass aus Fettsäuren kein Pyruvat gewonnen werden kann. Das bedeutet
wiederum: wir können zwar aus Zucker Fettsäuren synthetisieren aber nicht umgekehrt aus
Fettsäuren Zucker machen.
2. Der Citratzyklus – katabolisch betrachtet
Nun haben wir also unser Acetyl-CoA auf verschiedenen Wegen gewonnen und können uns dem
Citratzyklus widmen. Beginnen wir mit der rein katabolischen Funktion des Citratzyklus und gehen
später auf seine anderen Leistungen ein. Im Kleingedruckten befinden sich
Zusatzinformationen die nicht zum primären Verständnis einer Reaktion gebraucht werden. Sie
dienen aber einem besseren Verständnis biochemischer Zusammenhänge und sollten deshalb
nicht außeracht gelassen werden.
2.1 Kondensation: Acetyl-CoA + Oxalacetat ® Citrat
Die Acetylgruppe wird unter Wassereinlagerung mit Oxalacetat verbunden. Durch die Abspaltung
der Acetylgruppe liegt das Coenzym A wieder in freier Form vor und steht somit wieder für andere
Prozesse zur Verfügung. Katalysiert wird dieser stark exergone Prozess (freie Standardenthalpie G
o’ = -31,4 kJ/mol) von der Citrat-Synthase. Das Oxalacetat wird, wie im späteren Verlauf deutlich
zu erkennen ist, im Citratzyklus nicht verbraucht und kann diesen somit mehrmals hintereinander
durchlaufen. Es dient dem Prozess also nur als „Trägermolekül“ für die Acetylgruppe.
Synthasen wie die Citrat-Synthase verbrauchen bei ihrer Arbeit im Gegensatz zu Synthetasen kein
ATP.
Die erste Reaktion des Citratzyklus wird als eine Kondensation bezeichnet, d. h. zwei Moleküle
vereinigen sich zu einem größeren Molekül unter Abspaltung kleiner Moleküle wie z. B. Wasser.
Da das abgespaltene Coenzym A aber alles andere als klein ist, wird der Begriff der Kondensation
hier stark strapaziert.
4. Citratzyklus 20.12.2015 EuSaB e.V.
Acetyl-CoA reagiert unabhängig von der Stoffwechselsituation mit Oxalacetat zu Citrat. Dies liegt
daran, dass wie oben schon erwähnt Acetyl-CoA nicht selbstständig aus demMitochondrium
hinausgelangt, Citrat aber schon. Diesen Umstand nutzt das Acetyl-CoA aus, da es für die
Biosynthese von Lipiden im Zytoplasma gebraucht wird und somit das Mitochondrium verlassen
muss.
An dieser Stelle können also noch zwei entgegengesetzte Wege eingeschlagen werden: Abbau
oder Synthese.
Oxalacetat kann auch zur Aminosäurensynthese genutzt werden, darauf wird aber später noch
eingegangen.
2.2 Isomerisierung: Citrat ® Isocitrat
Für eine Oxidation ist Citrat ungeeignet, da die einzige Gruppe die potentiell dafür in Frage käme
als tertiärer Alkohol vorliegt. Von der
Oxidationsstraße wissen wir, dass nur primäre und sekundäre Alkohole weiter oxidiert werden
können. Also wandelt die Natur Citrat in Isocitrat - ein sekundärer Alkohol - um.
Zuerst wird enzymatisch per Dehydratisierung (Abspaltung von Wasser) das Zwischenprodukt cis-
Aconitat gebildet. Die tertiäre Hydroxy-Gruppe des Citrats wird dabei aufgelöst. Anschließend
sorgt das gleiche Enzym, die Aconitase per Hydratisierung (Einlagerung von Wasser) für eine neue
Hydroxy-Gruppe. Diesmal aber am zweiten C-Atom. Dieser sekundäre Alkohol kann nun weiter
oxidiert werden.
Beide Moleküle, Citrat und Isocitrat besitzen die gleiche Summenformel haben aber eine andere
Struktur. Man nennt diese Umwandlung deshalb eine Isomerisierung.
Isocitrat enthält im Gegensatz zum nicht-chiralen Citrat zwei chirale Zentren (C2 und C3) und
könnte somit in vier verschiedenen Formen auftreten. In unserem Körper wird aber nur 2R, 3S –
Isocitrat gebildet.
2.3 Oxidation und Decarboxylierung: Isocitrat ®a-Ketoglutarat
Nach der Isomerisierung kann nun die erste von insgesamt vier Oxidationsschritten des
Citratzyklus durchgeführt werden. Die Reaktion wird durch die Isocitrat-Dehydrogenase
katalysiert. Zuerst werden, wie der Name des Enzyms schon vermuten lässt, zwei Wasserstoff-
Atome abgespalten. Aus der Hydroxy-Gruppe am zweiten C-Atom wird eine Oxo-Gruppe (auch
Keto-Gruppe genannt).
Der Elektronen-Carrier NAD + nimmt nur eines der abgespaltenen H-Atome aber beide
Elektronen auf (= H - , ein Hydrid-Ion) und wird zu NADH. Übrig bleibt dann ein Proton ( H+)
welches frei in der Lösung schwimmt.
5. Citratzyklus 20.12.2015 EuSaB e.V.
Durch diesen ersten Teilschritt ist die instabile b-Ketosäure Oxalsuccinat entstanden. Noch an das
Enzym gebunden wird im zweiten Teilschritt ein Kohlendioxid entfernt (Decarboxylierung). Es
entsteht a-Ketoglutarat. Das a im Namen bedeutet, dass die Keto-Gruppe sich am ersten C-Atom
nach der Säure-Gruppe befindet.
NADH wird später in der Atmungskette für die ATP-Gewinnung verwendet. Ein Molekül NADH
liefert in der Atmungskette 2,5 Moleküle ATP.
a-Ketoglutarat kann zur Aminosäurensynthese genutzt werden, darauf wird später noch
eingegangen.
2.4 Oxidation und Decarboxylierung: a-Ketoglutarat ® Succinyl-CoA
Nun der zweite Oxidationsschritt des Citratzyklus. Er ähnelt dem Vorangegangenen sehr: Eine a-
Ketosäure wird oxidativ decarboxyliert und NAD+ fungiert wieder als Elektronenakzeptor für den
abgespaltenen Wasserstoff. Zusätzlich wird diesmal Energie in Form einer Thioesterbindung über
das Coenzym A gespeichert. Das Endprodukt ist dann Succinyl-CoA.
Katalysiert wird die Reaktion von der a-Ketoglutarat-Dehydrogenase.
a-Ketoglutarat-Dehydrogenase ist ein Enzymkomplex aus drei Enzymen. Es ähnelt im Aufbau und
Reaktionsablauf stark der Pyruvat-Dehydrogenase, welche die Umwandlung von Pyruvat in
Acetyl-CoA katalysiert.
2.5 Reaktion: Succinyl-CoA ® Succinat + CoA
Die energiereiche Thioester-Bindung wird durch das Enzym Succinyl-CoA-Synthetase (Succinat-
Thiokinase) gespalten. Die dabei entstehende Energie wird sofort genutzt um aus einem GDP-
Molekül durch Anlagerung eines anorganischen Phosphats GTP herzustellen. Dabei entsteht eine
ebenfalls energiereiche Phosphorsäure-Anhydridbindung.
6. Citratzyklus 20.12.2015 EuSaB e.V.
Das entstandene GTP ist mit ATP gleichzusetzen, da beide ohne Energieverlust mit dem Enzym
Nucleosid-Diphosphat-Kinase ineinander umgewandelt werden können.
Man nennt diesen Prozess, bei dem außerhalb der Atmungskette ein energiereiches
Nucleosidtriphosphat (ATP, GTP usw.) gebildet wird Seitenkettenphosphorylierung. Die Energie
die bei Umwandlung von Substraten in einer Stoffwechselkette frei wird, geht dadurch nicht als
Wärme verloren sondern wird durch Anlagerung einer Phosphatgruppe gespeichert. Anschließend
wird diese Phosphatgruppe samt Energie auf ein Nukleosid-Diphosphat wie ADP übertragen. Das
daraus entstandene ATP ist der Energielieferant schlechthin in unserem Körper.
2.6 Regeneration von Oxalacetat
Im Endeffekt muss für die Regeneration von Oxalacetat aus Succinat aus einer Methylengruppe
(CH2) eine Carbonylgruppe (C=O) entstehen. Dieser Vorgang wird in drei Teilschritten
durchgeführt: Oxidation – Hydratisierung – Oxidation. Hier kommt es also zu den letzten beiden
der insgesamt vier Oxidationen des Citratzyklus.
a) Oxidation: Succinat ® Fumarat
Die Succinat-Dehydrogenase entfernt unter der Ausbildung einer Doppelbindung
zwei Wasserstoff-Atome aus dem Succinat. Die dabei freiwerdende Energie
reicht nicht aus um ein Hydrid-Ion auf NAD+ zu übertragen. Deswegen weicht die
Natur auf einen anderen Elektronen-Akzeptor aus der schon mit weniger Energie
zufrieden ist, das FAD. FAD nimmt beide Wasserstoff-Atome auf und wird zu
FADH2. Da es weniger Energie brauch, wirft es in der Atmungskette auch nicht
soviel ATP ab wie NADH/H+ (1,5 statt 2,5 ATP pro Molekül ).
Die Succinat-Dehydrogenase, ist das einzige Enzym des Citratzyklus, welches
direkt in der inneren Mitochondrienmembran verankert ist. Sie ist das Bindeglied
zwischen Citratzyklus und Atmungskette und wird auch als Komplex II der
Atmungskette bezeichnet. FADH2 ist kovalent an dieses Enzym gebunden und
gibt beide Wasserstoff-Atome wieder direkt an dieses ab. Schon ist man
mittendrin in der Atmungskette.
b) Hydratisierung: Fumarat ® Malat
Die Fumarat-Hydratase (Fumarase) lagert Wasser an das Fumarat , dabei wird die
in a) gebildete Doppelbindung wieder aufgelöst.
Das Wasser wird genauer als eine Hydroxy-Gruppe und ein Wasserstoff addiert. Dadurch
entsteht ein chirales Zentrum, da nun eines der C-Atome über vier verschiedene
Substituenten verfügt. In unserem Körper entsteht nur die L-Isoform des Malats.
c) Oxidation: Malat ® Oxalacetat

Nun noch der letzte Schritt des Citratzyklus und der Kreis schließt sich. Die Malat-
Dehydrogenase spaltet zwei Wasserstoffe vom Malat ab. Dabei wird wieder NAD+ als
Elektronenakzeptor eingesetzt und es entsteht NADH/H+.
Oxalacetat hat also den Citratzyklus „überlebt“ und steht wieder als Trägermolekül für die
nächste Runde bereit.
Das Reaktionsgleichgewicht liegt eigentlich stark auf der Seite der Edukte, da die Änderung
der freien Standardenthalpie (D G O´ = + 29,7 kJ/mol) positiv ist. Die Reaktion wird aber
dadurch angetrieben, das ihre Produkte gleich weiterreagieren: Oxalacetat geht wieder in den
Citratzyklus und NADH in die Atmungskette. Somit kommt es selten zu einer Rückreaktion in
Richtung Malat.
3. Bilanz des Citratzyklus
Am Anfang standen zwei Kohlenstoff-Atome in Form von Acetat, die wir vollständig oxidieren
wollten um sie dann als Kohlendioxid abzuatmen. Auf dem Weg dieser C-Atome durch den
Citratzyklus gab es mehrere Schritte bei denen unser Organismus Energie gewinnen konnte:
Vier Oxidationsschritte bei denen jeweils zwei
Wasserstoff-Atome abgespalten wurden. Dreimal hat
die frei werdende Energie gereicht um NADH/H+ zu
bilden, einmal hat es nur für FADH2 gereicht.
Bei der Spaltung der Thioesterbindung des Succinyl-CoA
wurde über Seitenkettenphosphorylierung ein Molekül
GTP gewonnen.
Verbraucht wurden in dieser langen Prozesskette zwei Moleküle Wasser.
Nicht zu vergessen ist das NADH welches bei der Umwandlung von Pyruvat zu Acetyl-CoA
entsteht. Da es aber nicht direkt zum Citratzyklus gerechnet wird, taucht es in seiner
Endabrechnung auch nicht auf.
Unser Körper nutzt als „Energiewährung“ Nukleosidtriphosphate wie ATP. Im
Citratzyklus entsteht davon aber gerade mal ein Molekül direkt. Die restliche Energie liegt in Form
von reduzierten Elektronen-Carriern (NADH, FADH2) vor. Diese werden in der anschließenden

Atmungskette mit Sauerstoff oxidiert, dabei geben sie ihre H-Atome bzw. ihr Hydrid-Ion wieder ab
und stehen dem Citratzyklus für eine erneute Reduktion (Elektronenaufnahme) zur Verfügung.
Man erkennt hierbei, dass der Citratzyklus auf die aerobe Atmungskette angewiesen ist. Er kann
also im Gegensatz zur Glykolyse nicht auf anaerobe Arbeit umschalten.
Nun zur Bilanz:
Citratzyklus Atmungskette
3 x NADH ® 7,5 ATP
1 x FADH2 ® 1,5 ATP
1 x GTP ® 1 ATP

1 x Acetat ® 10 ATP
Insgesamt werfen die beiden C-Atome des Acetats über Citratzyklus und Atmungskette also 10
Moleküle ATP ab.
Mit Hilfe von radioaktiver Markierung wurde festgestellt, dass die C-Atome des Acetats, welches
in den Citratzyklus eintritt nicht identisch sind mit denen die den Citratzyklus als
KohlendioxidMoleküle verlassen. Es werden also nicht unbedingt die gleichen Kohlenstoffe
abgeatmet, die sich im Acetat befanden.
Verfolgt man den aeroben Abbau eines Glukose-Moleküls über Glykolyse ® Citratzyklus ®
Atmungskette so ergeben sich daraus satte 32 Moleküle ATP. Wenn man aber vergleicht, dass
unser Körper in Ruhe 40 Kg ATP pro Tag verbraucht relativiert sich dieser Wert wieder. Auch
interessant, bei einem zweistündigen Lauf verbraucht man ca. 60 Kg ATP.
4. Der Citratzyklus – anabolisch betrachtet
Nachdem wir gesehen haben welche Zwischenprodukte beim Abbau eines Acetats im Citratzyklus
entstehen, lenken wir unseren Blick nun auf die Biosynthesewege, die ebendiese Abbauprodukte
zum Aufbau neuer Substanzen nutzen.
9. Citratzyklus 20.12.2015 EuSaB e.V.
4.1 Acetyl-CoA: Die innere Mitochondrienmembran ist für das in der
Mitochondrien-Matrix gebildete AcetylCoA nicht passierbar. Durch die
Kondensation mit Oxalacetat zu Citrat kann nun aber ein
Citrat/MalatAntiport (Citrat raus, Malat rein) genutzt werden um das
Mitochondrium zu verlassen. Im Zytoplasma sorgt dann die ATP-
abhängige Citrat-Lyase die Aufspaltung zurück in Acetyl-CoA und
Oxalacetat. Das Oxalacetat wird über eine zytoplasmatische Malat-
Dehydrogenase in Malat umgewandelt und kann nun über den gleichen
Antiport wieder zurück ins Mitochondrium gelangen. Das Acetyl-CoA kann im Zytoplasma zur
FettsäurenSynthese und zur Herstellung von Isoprenoiden wie Cholesterin herangezogen
werden.
Wie weiter oben schon erwähnt findet der Fettsäurenabbau in den Mitochondrien, der
Fettsäurenaufbau hingegen im Zytoplasma statt. Diese strikte räumliche Trennung sorgt dafür
das Abbau und Synthese getrennt reguliert werden können und sich mit ihren
Zwischenprodukten nicht „in die Quere kommen“.
4.2Das a-Ketoglutarat dient als Grundbaustein für die Aminosäuren der
Glutamat-Familie. Dazu zählen
Glutamat, Glutamin, Arginin und Prolin. Durch Einlagerung von
reduziertem Stickstoff werden diese Aminosäuren unter
Energieaufwand hergestellt. Glutamat dient hierbei als Vorstufe für die anderen Aminosäuren
aus dieser Familie.
Es ist wichtig sich noch einmal vor Augen zuführen, dass die
Syntheseschritte auch rückwärts funktionieren und somit
beim Abbau von Aminosäuren der Glutamatfamilie a-
Ketoglutarat in den Citratzyklus einfließt.
4.3Succinyl-CoA dient als Ausgangsstoff für Porphyrine. Der
bekannteste Vertreter dieser Gruppe ist das Häm, welches wir z. B. für
die Sauerstoffanlagerung im Hämoglobin benötigen. Die Synthese des
Häms geschieht deshalb auch dort, wo rote Blutkörperchen gebildet
werden – im Knochenmark. (Ein geringer Teil des Häms wird auch in
der Leber gebildet).
4.4 Malat und Oxalacetat: Diese beiden Stoffe werden hier zusammen genannt, da Oxalacetat
das Mitochondrium nicht direkt verlässt sondern erst noch zu Malat reduziert wird.
Anschließend geht es über das sogenannte Malat-Shuttle rüber ins Zytoplasma. Dort sorgt eine
Malat-Dehydrogenase für die Rückwandlung in Oxalacetat.

Im Zytoplasma kann das Oxalacetat nun für zwei Synthesewege genutzt werden:
· Biosynthese der Aminsäuren aus der Aspartat-Familie (Oxalacetat ist genau wie a-Ketoglutarat
eine a-Ketosäure)
· Gluconeogenese: Durch die Phosphoenolpyruvat-Carboxykinase entsteht aus Oxalacetat
Phosphoenolpyruvat. Läuft meistens in der Leber und der Niere ab.
Der Großteil des für die Gluconeogenese benötigten Oxalacetats wird durch den
Aminosäureabbau geliefert. Entweder direkt in Form von Oxalacetat oder über Pyruvat, welches
dann im
Citratzyklus zu Oxalacetat carboxyliert wird.
Auch Lactat wird für die Gluconeogenese herangezogen. Es wird per Lactat-Dehydrogenase zu
Pyruvat.
In der Glykolyse wird aus Phosphoenolpyruvat Pyruvat – warum geht man diesen Schritt nicht
einfach rückwärts?
Bei der Glykolyse werden ein ATP gebildet und gleichzeitig noch ca. –30 kJ/mol an Energie
freigesetzt. Diese Energie ist verloren. Für den entgegengesetzten Prozess müssten also zwei ATP
aufgewandt werden. Bei der Umwandlung von Oxalacetat in Phosphoenolpyruvat wird nur ein
ATP benötigt, deswegen geht der Organismus den energiesparenden Umweg über die
Mitochondrien und den Citratzyklus.
5. AnaplerotischeReaktionen
Nachdem man den Citratzyklus auch anabolisch betrachtet hat, sei noch auf einen wichtigen
Aspekt hingewiesen. Wenn Zwischenprodukte des Citratzyklus für Biosynthesezwecke abgezwackt
werden, müssen diese auch wieder rasch aufgefüllt werden, um den Zyklus aufrecht zu erhalten.
Prozesse die einen Stoffwechselkreislauf wieder vervollständigen nennt man anaplerotisch (gr.:
auffüllen).
Wird beispielsweise viel Oxalacetat für die Biosynthese von Aminosäuren verbraucht, so kann
Pyruvat zur Neusynthese herangezogen werden. Der Prozess wird von der Pyruvat-Carboxylase
katalysiert. Dieses Enzym ist nur in Gegenwart von Acetyl-CoA aktiv, da eine hohe mitochondriale
Acetyl-CoA – Konzentration ein Zeichen dafür ist, das der Citratzyklus zu langsamläuft. Mit dem
neugewonnenen Oxalacetat wird dieser wieder beschleunigt. Wenn viel Pyruvat vorhanden ist
wird die Pyruvat-Carboxylase ebenfalls stimuliert.
Auch Fumarat kann anaplerotisch wirken, da es als „Abfallprodukt“ im Harnstoffzyklus entsteht
und in den Citratzyklus eingebaut wird.
Wichtig zu wissen ist, dass der Citratzyklus - da es sich um
einen Kreislauf handelt - an jeder beliebigen Stelle aufgefüllt
werden kann, egal welches Zwischenprodukt gerade für
Biosynthesen verbraucht wurde.

Die Regulation des Citratzyklus
Durch die bisherigen Ausführungen sollte klar geworden sein wie Komplex dieser Zyklus ist. Er
dient als Drehscheibe für viele katabole und anabole Stoffwechselprozesse und muss
dementsprechend genau reguliert werden.
Als Grundüberlegung der Stoffwechselregulation, kann man sich klar machen, dass hohe
Konzentrationen von Reaktionsprodukten und energiereichen Molekülen (z. B. ATP, NADH)
hemmend auf ein katabolisches Enzym wirken. Während eine hohe Edukte-Konzentration, ADP
und NAD+ aktivierend wirken.
Als Kontrollstellen eignen sich Enzyme, die eine stark exergone Reaktion katalysieren, da diese
Reaktion schwer bis nicht reversibel sind und deswegen auch nur durchgeführt werden sollten
wenn es wirklich nötig ist.
Der erste und wichtigste Kontrollpunkt ist die Pyruvat-Dehydrogenase:
Eine entscheidende Stelle, da die Umwandlung von Pyruvat in Acetyl-CoA für unseren Organismus
irreversibel ist.
Die stärkste Hemmung erfolgt über eine Kinase, welche die Pyruvat-DH phosphoryliert und
dadurch in einen inaktiven Zustand versetzt.
Wirkung auf die PDH-hemmende Kinase:
+ ATP, NADH, Acetyl-CoA -- ADP, NAD+,
Pyruvat, CoA
NADH und Acetyl-CoA wirken auch direkt allosterisch hemmend auf die Pyruvat-DH.
Die Aktivierung der Pyruvat-DH funktioniert genau umgekehrt. Aktiv ist dieses Enzym in seiner
nicht-phosphorylierten Form, also bedarf es einer Phosphatase.
Die Phosphatase wird nur von der mitochondrialen Calciumionen-Konzentration in ihrer Aktivität
gestärkt.
Der erhöhte CA2+-Spiegel wird durch a1-adrenerge Agonisten und Hormone (z. B. Vasopressin)
ausgelöst.
Insulin will den Glukosespiegel senken und kanalisiert Glukose in Richtung Pyruvat. Gleichzeitig
beschleunigt es die Pyruvat-Dehydrogenase.
Zwei weitere Kontrollpunkte sind die Isocitrat-Dehydrogenase und die a-Ketoglutarat-
Dehydrogenase.
Die Hemmung erfolgt bei beiden direkt über ATP und NADH. Zusätzlich wirkt auf die a-
Ketoglutarat-DH noch das von ihr katalysierte Reaktionsprodukt Succinyl-CoA hemmend.
Auf die Isocitrat-DH wirkt als einzige aktivierende Substanz ADP.

Beide Enzyme werden rein allosterisch (gr.: allos = fremd) beeinflußt. Das heißt eine Substanz
lagert sich an das Enzym an und verändert dessen Teritär- bzw Quartär-Struktur. Dieser
allosterische Effekt kann je nach Enzym und Substanz hemmend oder aktivierend wirken.
Kurzer Ausflug in die Klinik: Beriberi
Die Pyruvat-Dehydrogenase ist ähnlich aufgebaut wie die a-Ketoglutarat-DH (kommt später im
Citratzyklus) und die Transketolase des Pentosephosphatweges (ein weiterer Stoffwechselweg).
Alle drei sind große Enzymkomplexe, welche mehrer katalytischer Cofaktoren bedürfen um
arbeiten zu können.
Einer davon ist das Thiaminpyrophosphat. Wichtig für dieses CoEnzym ist - wie der Name schon
sagt - Thiamin (Vitamin B1). Kommt es wegen Fehl- oder Unterernährung zu einem Thiamin-
Mangel, sinkt die Enzymaktivität, die Reaktionen laufen seltener ab und es häufen sich die Edukte
an. Die durch die betroffenen Enzyme eigentlich verstoffwechselt werden sollten.
Im Klartext heißt das: Pyruvat- und a-Ketoglutarat-Spiegel sind bei Beriberi im Blut erhöht.
Folgen dieser erhöhten Werte sind: neurologischen Störungen und Veränderungen der
Herzaktivität. Durch Schädigung des peripheren
Nervensystems kommt es zu Gliederschmerzen, Muskelschwäche und Sensibilitätsstörungen der
Haut. Das Herz kann vergrößert sein und eine unzureichende Pumpleistung liefern. Warum ist
ausgerechnet das Nervensystem so stark davon betroffen? Da die Zellen des Nervensystems
vorwiegend Glykolyse zur Energiegewinnung betreiben sind sie besonders von einem Ausfall der
Pyruvat-DH betroffen. Pyruvat ist das Produkt der aeroben Glykolyse und hemmt die Glykolyse
vielleicht (VERMUTUNG!) über eine negative Feedbackschleife, wenn es nicht über die Pyruvat-DH
weiter verarbeitet wird.

Beriberi tritt vor allemin asiatischen Ländern auf, da das dortige Grundnahrungsmittel Reis arm
an Thiamin ist. Auch unterernährte Alkoholiker können von einem Thiaminmangel betroffen sein.
Zu ähnlichen Symptomen wie bei Beriberi kann es auch durch Arsen- oder
Quecksilbervergiftungen kommen. Beide Stoffe greifen ebenfalls die Pyruvat-DH an.
