modifications and nucleotide
analogs could be accepted by
the RSV RNA polymerase”
5. Materiales y métodos: RNA recombinante del
RSV
Definición
• Es una molécula de RNA artificial
creada in vitro al unirle un
fragmento de RNA exógeno.
Para qué se usó
• Para lograr formar el complejo de la

6. Materiales y métodos: Elongación
Definición
Proceso dentro de la transcripción
en el cual después de ubicarse la
RNA polimerasa en el sitio
promotor se inicia la
incorporación de rNTP y se forman
enlaces fosfodiester.
Para qué se realizó
Para cuantificar la habilidad de los
nucleótidos análogos y los primer
para realizar este proceso. Se
utilizó: el molde de RNA, el primer,
el NTP nativo o análogo y un
nucleótido marcador.
7. Materiales y métodos:Autorradiografía
Definición
• Proceso por el cual se sustituyen
átomos por complejos
radioactivos como el H3 o I, de
esta manera se interrumpe los
procesos biológicos y se fija, así
la partícula queda con el isótopo
radioactivo y se puede leer con
una película de fotografía.

¿Para qué se uso?
• Para observar los productos de
las reacciones de elongación.

8. Materiales y métodos: Oligonucleótidos
Definición
• Es un fragamento de DNA o RNA
que presenta una alta densidad
en una superficie pequeña. Son
solo de 13 a 25 nucleótidos.
Tomado de:https://www.news-medical.net/life-sciences/What-is-an-
Oligonucleotide-(Spanish).aspx
¿Para qué se usó?
Ya que resultaba imposible purificar
un molde de N-RNA, los
oligonucleótidos sirvieron como
molde donde se incubó el complejo
RSV L-P para llevar a cabo la síntesis
del RSV RNA.

13. Discussion
Author Mencioned by the author Yes or No
Cressey et al., 2018; Noton et al.,
2012; Tremaglioetal.,2013
“In its native state, the RSV
polymerase does not use a primer
to initiate RNA synthesis”
Tchesnokov et al., 2018
“It was recently reported that a
5′phosphate group on the ACGC
primer was essential for templated
primer elongation on an 11-mer
template derived from the RSV
genomic promoter”
RehaKrantzetal.,2011
The polymerase could incorporate
the intrinsically fluorescent purine
analog 2-aminoPTP in place of ATP
but not in place of GTP even though
research with DNA polymerases has
shown that 2-aminopurine is able
to pair with cytidine
14. Conclusions
The study of the functioning of the
polymerase of the RSV shows a
development in the
understanding of the ways that
this virus Works, this is an
essencial point for posible future
treatments.
The RSV´s polymerase
elongation process study
demonstrated that primers are
requiered (even the ones that
have bulky chains), also a
polymerase without mutation
and that this protein
(polymerase)can accept
different nucleotide analogs, but
always conserving the law about
the complementary bases.
