Abstract— Taxus Chinensis var. mairei is a valuable plant species for timber and taxoids isolated from this species are very important compounds that are used for cancer treatment. Although chemical investigation on T. chinensis var. mairei are popular, functional identification of genes isolated from this species is rare. In this investigation, we have isolated TCAP3 gene and analyzed its expression pattern in different tissue and developmental stages through Real time-PCR; then we transformed this gene into Arabidopsis and analyzed its function. Our results demonstrated that its cDNA contains 846 bp bases (coding 197 amino acids) constituted by four typical domains, M, I, K, C with conserved motif, Phylogenetic analysis showed that TCAP3 is more ancient than angiosperm B class genes. Alignment of protein sequence demonstrated the conserved motifs, which illustrated that TCAP3 belongs to gymnosperm Gymno B class MADS-box genes with PI-derived, on C-teminal, which is similar structure to the Gymno B class MADS-box genes that they share the same B class gene specific conserved motif. Expression analysis of TCAP3 in different tissue showed that it only expression in male strobilus, not in leaf, bud and female strobilus at different developmental stages. We divided the stages according to paraffin sections of male strobilus. The results indicated that TCAP3 expresses dynamically along with the male strobilus. Heterologous expression of TCAP3 in Arabidopsis demonstrated that TCAP3 was involved in flower, especially the filaments morphological development.

Tomato (Solanum lycopersicum L.) is most important Solanacous vegetable grown worldwide for its edible fruits. Various marker techniques have been successfully applied, either individually or in combination to study the genetic diversity of this crop. A Study to assess the usefulness of different markers system for analyzing the genetic diversity and relation between different varieties and to find out correlation between marker systems revealed that all tested tomato cultivars could be differentiated from each other based on either morphological/protein/RAPD markers individually, and can be applied for grouping of cultivars, pedigree analysis and genetic diversity analysis. However, markers system used in this study showed variations in understanding the genetic relation between studied varieties.

In the present investigation, based on the seven rice putative candidate iron transporter genes, novel TRAP markers were developed. These markers were successfully employed in the molecular diversity study among 30 rice genotypes representing improved rice cultivars and land races with varied grain iron content (7.38 - 30.58 ppm). Totally, thirty TRAP primer combinations were screened, which generated 703 bands out of which 654 were polymorphic (93%) with an average of 21.8 bands per primer combination. The average polymorphic information content (PIC) values ranged from 0.09 (Osysl4b+ME05) to 0.25 (Osnramp5c+ME05, Osnramp1b+ME02 and Osysl4a +ME02). Gene diversity (H ˆ ) ranged from 0.10 (Osysl4b+ME05) to 0.31 (Osnramp1b + ME02 and Osysl4a +ME02). The Jaccard dissimilarity ranged from 0.15 to 0.52, explaining 37% of genetic variation (Table 4). Grouping of genotypes based on UPGMA and principal coordinate analysis (PCoA) were found comparable and grouping of genotypes into a different cluster was found mainly on the basis of pedigree relationships. TRAP markers revealed well resolved relationships among rice genotypes. The information generated from this study will helps to select parental combinations for breeding high iron content rice varieties.

Inter-simple sequence repeat (ISSR) analysis was performed in seven accessions of Solanum xanthocarpum Schard. and Wendl. of Assam to evaluate the applicability of this analysis for assessing the intraspecific variation. The value of similarity indices ranged from 0.375 to 0.125. The similarity result indicates the presence of high level of genetic diversity among the accessions of Solanum xanthocarpum Schard. and Wendl. UPGMA cluster analysis revealed clear grouping among the populations. The primers showed abilities in detecting genetic diversity across wild accessions of Solanum xanthocarpum Schard. and Wendl. Thus, ISSR-PCR technology can be used to study genetic variation and genetic relationships in the genus Solanum xanthocarpum Schard. and Wendl. Early generation selection in segregating peanut populations used markers linked to high oleic acid, rust and late leaf spot (LLS) resistance coupled with near-infrared reflectance spectroscopy (NIRS) for high oleic trait for selection decisions. Techniques such as rapid generation advancement, recycling of elite lines as parents, and early generation and multi-location testing in target sites resulted in an enhanced genetic gain for the high oleic trait. The significant outcomes are combining early maturity with foliar fungal disease resistance, and agronomically superior high oleic lines. For the 100-seed mass, a genetic gain of 17 g was achieved, from 35 g in 2015 to 52 g in 2017, through recycling of elite lines along with directional selection for seed size in the high oleic breeding pipeline. Superior performing high oleic as well as early maturing rust and LLS resistant lines are under national testing for release in India.

Random Amplified Polymorphic DNA analysis is an excellent tool as a biomarker in determining genetic purity and estimating genetic diversity in plant species. Seven species of Ficus genus that are locally available in Hyderabad such as Ficus benjamina, Ficus microcarpa, Ficus elastica, Ficus binnebdjkii, Ficus benghalensis, Ficus religiosa and Ficus carica. Those ficus cultivars were then analysed by using three oligonucletide primer Z13 (5’GACTAAGCCC 3’), Z17 (5’ CCTTCCCACT 3’) and Z18 (5’AGGGTCTGTG 3’). A total of 340 amplified fragments, in which 212 (62.4%) were polymorphic fragments. The number of polymorphic bands scored per primer ranged from 9 (primer Z13) to 25 (primer Z18). Sixty out of 340 RAPDPCR fragments were found to be useful as cultivar specific markers. Genetic similarities among the seven Ficus cultivars were estimated according to RAPD and cultivar distribution on the concensus tree according to banding pattern of RAPD. Results of combined data exhibited that Ficus benghalensis and Ficus elastica are related cultivars with highest similarity index. On the other hand Ficus benghalensis and Ficus benjamina are two distant related species with low similarity index.

Ginger (Zingiber officinale) rhizomes have been widely used as a spice and flavoring agent in foods and beverages in Bangladesh as well as in all over the world for its economical and medicinal values. The present investigation was undertaken for the assessment of 13 local ginger genotypes collected from different region of Bangladesh through 7 RAPD primers. Genomic DNA was extracted from ginger genotypes using CTAB method. A total of 34 distinct and differential amplification bands ranging from 150-1200 bp were observed with an average of 1.14 polymorphic bands per primer. The overall gene diversity was detected 0.8052 and the value of PIC was detected 0.7532. The RAPD marker generate enough polymorphism for possible use in diversity studies through cluster analysis and principal component analysis (PCA). PCA classified 13 ginger genotypes into four groups and showed in two dimensional scatter plot. The genetic similarity coefficients among genotypes ranged from 0.103 to 0.654. Cluster analysis based on Jaccard’s similarity-coefficient using UPGMA grouped the genotypes into two clusters: Cluster A and Cluster B. The cluster ‘A’ had only one genotype Kaptai local and the second cluster ‘B’ had rest of twelve genotypes. The prevalence level of polymorphism in the local genotypes of ginger will help to breeders for ginger improvement program.

Studying of genetic relationships among Aegilops L. species is very important for broadening the cultivated wheat genepool, and monitoring genetic erosion, because the genus Aegilops includes the wild relatives of cultivated wheat which contain numerous unique alleles that are absent in modern wheat cultivars and it can contribute to broaden the genetic base of wheat and improve yield, quality and resistance to biotic and abiotic stresses of wheat. The use of molecular markers, revealing polymorphism at the DNA level, has been playing an increasing part in plant biotechnology and their genetics studies. There are different types of markers, morphological, biochemical and DNA based molecular markers. These DNA-based markers based on PCR (RAPD, AFLP, SSR, ISSR, IRAP), amongst others, the microsatellite DNA marker has been the most widely used, due to its easy use by simple PCR, followed by a denaturing gel electrophoresis for allele size determination, and to the high degree of information provided by its large number of alleles per locus. Day by day development of such new and specific types of markers makes their importance in understanding the genomic variability and the diversity between the same as well as different species of the plants. In this review, we will discuss about genetic variability and phylogenetic relationships studies of Aegilops L. using some molecular markers, with theirs Advantages, and disadvantages.
