COMPARISON BETWEEN REPLICATION OF
PROKARYOTES AND EUKARYOTES
DNA Replication in
Prokaryotes
• In prokaryotes replication
start from ori c site known
as Origin of replication
site.
DNA Replication in
Eukaryotes
• The genome of
Eukaryotes is so much
large so its speed is very
slow that is 50 nucleotide
per second. And it has
many points from where
replication is started. It
has many origin of
replication.

Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
2
• It has small genome so
therefore.
• After each 30,000 or
300,000 nucleotides, there
is a new origin of replication
site.

3
• Ori c site. • Those sequences from
where replication starts is
called ARS site
Autonomous Replication
Site

• . Here DNA-A protein attach
with ori c site, which is
single protein.
• Here at ARS, attach a
multiple complexes of
group of protein called
origin recognition
complexes ORC: it is a
multiprotien complexes.
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
5
• Here DNA-C protein carry
the the DNA-B protein.
• Here a multiprotiens do this
work. These are
monochromosomes
maintenance protein (
MCM6, mco)
• These act as DNA-B protein
and cell division cycle6 or (
CDC6). CDT act as DNA-c
protein there are 2
multiprotien complexes
which act as DNA-C protein.
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
6
when DNA-C protein has
done its work, topoisomerase
come to attach on both sides.
Same is the case with,
topoisomerase 1 or
toporoisomerase2 attach on
both sides.
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
7
• Here SSBP attach with the
strand of DNA and keep
them in unwind protein, so
that they can’t rewind again.
• . RNA premise attach the
primer.
• Replication proteins a “RPA”
which is a multiprotien
complex. It stops to rewind
of nucleotides of eukukarytic
DNA.

Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
8
• Here DNA polymerase1.
• DNA polymerase2.
• DNA polymerase3.
Involve in replication
• Here DNA polymerase
alpha
• DNA polymerase delta and
• DNA polymerase epsilon.
Involve in replication
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
9
• All RNA primase makes the
primer.
• DNA polymerase alpha
makes the primer.
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
10
• DNA polimarse3 is involved
in main replication process.
• DNA polymers Delta carry
out the main replication.
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
11
• Here DNA polimarse1 or
DNA polimarse2 carry out
the proof reading.
• DNA polymerase epsilon do
the poof reading.
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
12
• Speed is 1000 nucleotide
per second.
• Replication speed is 50
nucleotide per second
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
mujahid.hussain7877@gmail.com
13
• Terminating proteins are
involved or Tus-Ter
complexes are present at the
end of strands.
17. Replication is bidirectional.
• Here long adenine chains ( of
100-200 nucleotide) called
talomess at the end of
strands.
15. Replication is also
bidirectional.
8/2/2017
Mujahid Hussain(M.Phil Botany) Department of Botany, UOS,
