5. About MPT64
 The MPT64 antigen is the secretory
protein (24 kDa) that is one of the
secreted major antigens from
tuberculosis bacteria,
 MPT64 has been shown to differentiate
the MTC from other bacterial species

8. Test Principle
 Sensit TB Ag MPT64 Rapid Test works on chromatographic
immunoassay. B
 Basic components of test strip includes:
a) Conjugate pad, which contains Monoclonal Anti-MPT64 antibody,
colloidal gold conjugated;
b) a nitrocellulose membrane strip containing two lines
a) T: Monoclonal Anti-MPT64 antibody and
b) C: Goat Anti Mouse.

9. Sample Preparation
 Liquid Cultures: One drop of liquid culture can
be directly added to the sample well of the test
device.
 If the culture is too turbid, adjust the opacity
with McFarland No 1 using PBS.
 · Solid cultures: Pick 3-4 colonies from solid
culture and suspend in 200 ul of Assay buffer.
 Treat the specimen as infectious and handle with standard
biosafety measures.

10. Test Procedure
 Take out the test card from the foil pouch and place it
on a horizontal surface.
 Add 25 ul (using pipette)/one drop (using dropper
provided) of the liquid or solid culture (processed as
above) to the sample well, followed by the addition of
one drop of assay diluent.
 Wait for 20 minutes and interpret results.
 The result is considered invalid after 30 minutes.
 All results where control band does not appear are
considered invalid.
