Genome size of a target species is a crucial consideration of any comparative genomic analysis. In complete genome sequencing projects, amount of DNA can be expected to be directly proportional to both financial and laboratory costs (Gregory, 2005). Among amphibians, genome size fluctuate more widely than in any other group of vertebrates (130-fold in contrast to 2-fold in birds or 5-fold in reptiles and mammals). We are interested in study this huge variation in amphibian genome size. “Why some species have larger genomes than others?” “Is genome size related to evolutionary history?” or “Is this variation consequence of habitat adaptation?” are some of questions we are looking to answer. Classical methods for estimating genome size employ DNA-specific fluorescent dyes in flow cytometry analysis (Hare et al. 2011), or absorption cytometry after Feulgen staining (Voglmayr & Greilhuber, 1998). However, these methods require the use of fresh non-processed tissues, and so genome size can’t be estimated on museum samples (Bainard et al. 2011). Our goal is to estimate genome size across all amphibians using a real-time PCR-based method (Wilhelm et al. 2003) with minor modifications. This method is based on the absolute quantification of a single copy gene in a genomic DNA sample by real-time PCR (qPCR). Through this method, the ‘C-value’ (i.e., mass of DNA per haploid genome) is calculated from the ratio of the mass of template DNA and the copy number of the target sequence determined by qPCR. We have already designed and preliminary tested candidate primers for real-time amplification of single-copy genes across amphibians (frogs, newts, salamanders and caecilians). Our preliminary results show that this method can be successfully applied when fresh tissue is not available. It is however well known that amphibian have huge proportion of inhibitors that could affect the qPCR success (Kosch & Summers, 2013). For this reason, we would like to verify our initial findings using different DNA extraction methodologies and further loci.

In BC’s nearly-decade-old “new” curriculum, the curricular competencies describe the processes that students are expected to develop in areas of learning such as mathematics. They reflect the “Do” in the “Know-Do-Understand” model. Under the “Communicating” header falls the curricular competency “Explain and justify mathematical ideas and decisions.” Note that it contains two processes: “Explain mathematical ideas” and “Justify mathematical decisions.” I have broken it down into its separate parts in order to understand--or reveal--its meaning. The first part is commonplace in classrooms. By now, BC math teachers—and students—understand that “Explain mathematical ideas” means more than “Show your work.” Teachers consistently ask “What did you do?” and “How do you know?” This process is about retelling, not just of steps but of thinking. The second part happens less frequently. Think back to the last time that you observed a student make—a necessary precursor to justify—a mathematical decision. “Justify” is about defending. Like “explain,” it involves reasoning; unlike “explain,” it also involves opinion and debate. In order to reinterpret the curricular competency “Explain and justify mathematical ideas and decisions,” I will continue to take apart its constituent part “Justify mathematical decisions” and carefully examine the term “mathematical decisions.” What, exactly, is a “mathematical decision”? Below, I will categorize answers to this question. These categories, and the provided examples, may help to suggest new opportunities for students to justify.
