  2. 2. Human Dermal Fibroblasts Human dermal fibroblasts (HDFs) in the skin are derived from mesenchymal stem cells (MSCs) during embryonic development. HDFs are abundant in the human body, they contribute to the formation of connective tissue and have multi-directional differentiation potential. When the body is damaged, fibroblasts are activated to participate in the production of the extracellular matrix and repair damaged tissues. 2 INTRODUCTION
  3. 3. Vascular Endothelial Cells It is the type of flat cell that lines the inside of all blood vessels and is in permanent contact with the blood. They have antithrombotic functions, selective permeability, modulation of blood flow and vascular resistance, regulation of immune responses, hormone synthesis, and lipoprotein modification. 3 INTRODUCTION
  4. 4. Identification of a new way to induce differentiation of dermal fibroblasts into vascular endothelial cells 4 OBJECTIVE
  5. 5. Western blot analysis 5 METHODS Quantitative real-time PCR It is a technique for identifying specific proteins in a complex mixture of proteins. Proteins are separated by electrophoresis and then incubated with antibodies that react with the protein of interest. It is a test to amplify a specific sequence of genetic material and see how much it was amplified to see the expression of the gene They used it to see the level of expression of the markers and genes of the HDF and VEC It was used to see the level of proteins of the HDF and VEC.
  6. 6. Immunofluorescence microscopy 6 METHODS Flow cytometry It is a complex technology that allows to analyze a cell population providing a wide variety of parameters, which can be correlated and displayed in different formats. It is an immunolabeling technique that uses antibodies chemically linked to a fluorescent substance to demonstrate the presence of a certain molecule. The microscope is then used to analyze images individually or overlaid. It was used to see the level of proteins of the HDF and VEC. It was used to see the level of VEC´s proteins and VEC´S gene expression after treatment of HDF.

DISCUSSION AUTHOR WHAT THEY SAID AGREE / DISAGREE Li L and Nourse MB (37-38) The VEGF signaling pathway plays a regulatory role in the differentiation of endothelial progenitor cells into VECs Agree Kemp SS and Bowers SLK(39- 40) PDGF signaling pathway also plays an important role in the diferentiation of endothelial progenitor cells into mature endothelial cells Agree Kano MR (41) VEGF and FGF-2 synergistically activate the endogenous PDGFB-PDGFRβ signaling pathway Agree
  10. 10. 10 CONCLUSIONS Thanks to the different techniques in molecular biology, great advances can be made that could allow new medical treatments in the future, as in this case, an alternative to repair damaged blood vessels and prevent ischemic processes. The understanding of molecular processes in cells allows scientific progress to propose new ideas for medical intervention in different fields, such as cell therapy in tissue recovery, cancer prevention, treatments for chronic diseases, etc.
