Abstract:
Purpose:This research try to build a HPLC method to detect and measure colchicine in the
blood of rats which is given orally with colchicine micro pellet.

Method: (1)The colchicine mirco pellet is made by fluidized bed. (2)The preparation of blood
sample:Take 5 drops blood from the rim of rat’s eye, centrifuge (4500R,10min), separate 50ul
upper liquid, add 0.3ml ethyl acetate, shake with
hand(20min) , centrifuge ( 3000R , 10min ) , separate 0.2ml upper liquid , blow with
nitrogen,add 1ml solution in reminder. Recovery of extraction: 30.5%,rsd:4.8% (3)The
method for giving micro pellet to rats: we temporarlly decide to use the drivepipe to tamp.
(4)The condition of HPLC: Chromatographic column: Kromasil 100-5C18 ( 250x4.6mm
E59392 ) ; Flow part: 45 % methanol + 55%H2O; Flow rate:1mL / min; Temperture of
column:20°C; Volum of sample:100ul; Wavelength of detect:254nm; solution: 10%methanol+
90%H2O; Linear scale: 10ng/ml~1ug/ml,y = 14954x - 1811.8,R² = 0.99999; Limit of
detect: 1ng; Limit of measure:5ng; recovery test(add sample): 80%group : 100.2% ;
100%group:98.7%;120%group:98.1%;rsd:1.09%.
Result: This method theoretically can be used in detection and mesurement for the blood of rats
given orally with big dose colchicine(, although I can not provide some experiment date to
verify it since the time of my internship is limited.
