  1. 1. Calcitonin • Peptide hormone • Parafollicular cells or clear (C) cells of thyroid gland synthesize and secrete calcitonin • Calcitonin is stored in the secretory vesicles
  2. 2. Stimulus of secretion of calcitonin • High serum Ca2+ levels High serum Ca2+ level Calcitonin release Acts on bones and kidney Decrease serum Ca2+ level
  3. 3. Physiological actions • On bone- osteoclast possess receptors for calcitonin • Inhibits bone resorption • On kidneys- decrease Ca2+ and phosphate reabsorption • Calcitonin levels are increased during pregnancy in order to prevent the bone loss in mother (bone formation and lactation are major Ca2+ draining events)
  4. 4. Mechanism of action of calcitonin • Calcitonin binds to GPCR which may activate either adenylyl cyclase or phospholipase C • Activation of protein kinase C and increase intracellular Ca2+ concentration • Release of calcitonin containing vesicles via exocytosis.
