Master’s Thesis: “Allelic Polymorphism Analysis of HLA Gene in HCC during Late HBV Infection”.
Thesis Description & Scope: This study focused to find out the HLA polymorphism and association of
alleles with the susceptibility to chronic Hepatitis B infection followed by the onset of Hepatocellular
carcinoma (HCC)
Objective: HLA typing of HBV-HCC patients was performed by SSP PCR method using certain allele
specific primers having potential effect in the chronic inflammation. Immunoglobulin & complement
levels were studied to check the chronic effect.
Conclusion: The study concluded that the presence of DQB10302, B0702 and DRB1404 alleles in the
HLA gene if found in a patient suffering of HBV infection, then the chronicity of the condition may lead to
HCC.

Major Project: “Construction of T-A Cloning Vector for PCR products.
Project Description & Scope: The T-A cloning method designed to facilitate the cloning of PCR
products having 3ʹA overhang, is easily converted to a universal cloning method.
Objective & result: In our study, Insertion of the oligos containing two tandemly arranged XcmI
restriction sites into pUC19
vector in between the EcoRI and Hind III restriction sites followed by digesting
with Xcm I restriction enzyme results in vector with T overhangs. The use of a linear “T-vector” which has
single 3'-T overhangs on both ends allows direct, high-efficiency cloning of PCR products, facilitated by
complementarity between the PCR product 3'-A overhangs and vector 3'-T overhangs.
