2. Background 1: food safe DIY culture
In past experiments, “Green Dakara” sports drink could be blended with
DMEM up to 60% without losing cell culture efficacy.

However, all ingredients must be replaced with commonly available
materials to enable kitchens DIY cell culture experiments.
※Research grade reagents = Not approved as food and not commonly available.
Food or food additives that can be ingested (within limits) are commonly available.
Basal media (DMEM, L15) was prepared entirely from commonly
available materials and a DIY cell culture was demonstrated.
3. Background 2: composition of DMEM and L15
The biggest difference between DMEM and L15 is in the minerals.
Other ingredients were assumed common.
Precisely, DMEM and L15 have different amino acid compositions and DMEM uses glucose while L15 uses galactose

General strategy for preparation
・Prepare 1X concentrated solution for salts, glucose, and amino acid, for which large quantity is needed.
・Prepare 20X solution for vitamin B’s and other minerals, for which only trace amount is needed.
・Prepare phosphates from phosphoric acid and mineral salts.
DMEM:
CaCl2 0.50g
MgSO4 0.25g
baking soda
0.19g
Phosphoric acid
diluted 10X 1.4ml
L15:
CaCl2 0.28g
MgSO4 0.20g
MgCl2 0.19g
K2CO3 0.06g
Phosphoric acid
diluted 10X 1.4ml
Mix 1X solution and 20x solution by 19:1, add glucose at 4.5g/L and bon apetit
8. Scenes from the experiment
・Filtration is needed, as the tablet excipients need to be removed.
・Wait until the suspension settles to avoid clogging the filter paper.
