4. Materials and Methods for AFM Adhesion Measurements
4.1. Materials
Paraformaldehyde (4% in 0.1 M Phosphate Buffer Saline), 1 Phosphate Buffered Saline (PBS) and
Muscovite Mica (V–1 Quality) were purchased from Electron Microscopy Sciences (Hatfield, PA).
Coverslips (No.1.5 thickness) were purchased from MatTek Corporation (Ashland, MA). MDA–
MB–231 and Hs578Bst cell lines, growth media (L–15 fand 46–X, respectively), and media
supplements (penicillin/streptomycin and fetal bovine serum) were purchased from American Type
Culture Collection (ATCC, Manassas, VA). Uncoated n–doped silicon AFM probes (MPP–31100)
were purchased from Bruker (Billerica, MA).

The breast cancer cells were incubated at 37 °C under normal atmospheric pressure, while the
normal breast cancer cells were incubated at 37 °C in environments with 5% CO2 levels,
respectively. The MDA–MB–231 and Hs578bst cells were grown respectively in L–15 or 46–X
base media. These were supplemented with 100 I.U./ml penicillin/100 μg/ml streptomycin and 10%
FBS. The cells were first rinsed with 1X PBS and then fixed with 4 wt% paraformaldehyde solution
for 10 mins at room temperature. They were then rinsed with PBS solution for three more times,
before a final rinse with purified water. Finally, the samples were dried in a vacuum desiccator for at
least 5 hours.
4.4. AFM tip Characterization
An FEI Quanta 200 FEG Environmental–SEM (FEI, Hillsboro, OR) was used to view the AFM tips
before and after coating. This was done to determine whether or not the AFM tips were successfully
dip–coated. The tips were also viewed in the ESEM after the pull–off tests. This was done to ensure
that no coating was lost during the AFM pull–off.
4.5. Force–Displacement Measurements
All of the AFM experiments were performed in a Dimension NanoMan AFM (Bruker, Billerica,
MA). This was done under ambient conditions at room temperature of 24 °C and a relative humidity

behind mixing primary colors to make secondary ones, and had even experimented with tertiary
colors on occasion. I even developed an innocent curiosity in the works of Leonardo da Vinci as I
grew older, but still maintained a blasé approach towards art in general. Since participating in Art
Appreciation at DCCC, my views on that matter have changed. I have gained a new fondness for the
seemingly trivial things in life and have since looked at them differently than I did before I signed
up for the class. Upon visiting the Reynolda House this weekend with a fellow classmate to work on
our first outside–of–class writing assignment, this adapted outlook was especially apparent. The two
of us wandered about the museum at a leisurely pace, and while I was already vaguely familiar with
the layout from a previous visit as a little girl, three oil paintings from the Victorian era caught my
eye. While they share an initial similarity with each other in their naturistic settings, they also differ
in a myriad of ways. The first piece of art that I will be analyzing is The Old Hunting Grounds, an
oil painting created by Worthington Whittredge in 1864. Whittredge largely limits his color pallete

comprehension by experimenting with optics that have different characteristics. Does one axis truly
get inverted when viewing an object through a lens? How significant of an effect does human error
have in calculating reflection (if any)? How much does magnification blur an image viewed through
an optic? How much does the angle of light deflect from a change of medium?
PROCEDURE: First, the law of reflection was examined. The law was verified by observing the
reflected image of a place marker, marking where the reflection appeared to line up with another
place marker, and then subsequently drawing a line along the line of sight for the pins. The angle
was then measured and verified through equivalency, as if the law of reflection is true then the
interior and exterior angles of the setup, with a line normal to the mirror's plane drawn in the center.

Part A– Microscope Parts
Scanning objective – These lenes start out at 4X, which mean 4 times 10 magnification, making the
lense a 40 magnification. You always want to start out on the smallest lens, get your specimen in
focus, and then move to the larger scanning objective lens.
Fine adjustment – The fine adjustment is going to be a knob that does not move the stage up and
down, but instead will bring your specimen into focus. When moving the scanning objective lenses,
you will always use the fine adjustment instead of the coarse adjustment to refocus the specimen.
Coarse adjustment– The coarse adjustment is the knob that will move the stage up and down. You
will use this to find the specimen, and then use the fine adjustment to focus.
Mechanical stage – The mechanical stage is the part of the microscope that you will put your slide
on. It holds the slide and enables it to move around.
Ocular Lens – The ocular lens is the lens you look in closet too your eyes. It helps magnify the
specimen as well.

can be seen as the sense of denial that takes place in a child's mind that is not allowed to openly
express herself, her fears, her anxieties, and her hopes. She allows the doll to represent her honest

Our night sky, always seems to be sparkling at night, due to stars. Stars are luminous ball of gas,
mostly hydrogen and helium, held together by its own gravity. Our night sky is filled with groupings
of stars creating a galaxy, or also known as The Milky Way. The Milky Way, is one of many
galaxies in a Local Group, containing between 100– 400 billion stars. Observing these stars with the

Depth of field is an element in cinematography that means a range of distance between objects and
how they are being shot in sharp focus. Depth controls the perspective relations by depicting what
planes will be in focus. There is also a director's decision to choose a selective view. It is used to
focus on only one plane and the others being blurred. Selective focusing automatically makes us pay
attention to one area equally visibly but not all being in sharp view. It also examines deep focus and
racking focus. Deep focus is a tight shot of an object; it's a shorter lens and offers better lighting to
be focused on. Attention is on several areas all being visible. Racking focus also known as "pulling
focus". The point of this focus is to transfers the attention from the foreground and the background.
It overall makes one action blurred and another in a sharp focus.
In the film Hurt Locker depth of field is very present. The clip I chose to give the example is

AAcademy Ratio Of 1.37
Prenom Carmen uses a academy ratio of 1.37:1. Starting with the first shot, it begins with a man,
named Uncle Jean, in the hospital staring out of the window. Godard's use of a middle lense, a
straight–on camera angle along with high key lighting all made the visuals seem somewhat muted.
The colors are all "soft" or "subdued" with no other prominent lighting effects in this first shot. Yet
while the visuals does not necessarily capture my attention, Godard's use of sound and framing
certainly does.
Godard uses framing and sound in conjunction to switch between offscreen and onscreen diegetic
sounds. Starting with the beginning of the first shot, the incredibly loud knock on the door
(offscreen diegetic) and Uncle Jean yelling, "Where am I?" (onscreen diegetic) as he moves from
the right of the frame to the center. The image size of the first scene is a medium long shot as I
noticed that only the knees and up are shown. I then hear two offscreen diegetic sounds of a woman
laughing with her high heels walking towards the frame. As the woman finally walks into the frame
and identifies herself as "Carmen" she bends down and kisses the man. Carmen then kisses
(onscreen diegetic) Uncle Jean, the sound of the kiss is also noticeably loud.
Carmen begins to walk out of frame and when she is out of frame, the non diegetic score kicks in
and then fades away for a brief moment to let Carmen and Uncle Jean talk. All of this happens as
Uncle Jean stays in the middle of the frame.

many ways. This artwork takes up almost the whole canvas. The horses on the edge look like they
were maybe even squeezed in. All of the characters in this artwork look to be congregated to the
middle of the painting, besides the villages in the distance. Linear perspective is used in the
placement of the villages in the background and in all the men and horses, which are grouped in the
middle of the screen. Looking at the features in the painting I notice the use of overlapping and
vertical placement, which both imply depth. The horses overlap each other as well as the gentlemen
and other elements of the painting. The brown horse's head overlaps the black one hiding its mouth.
The villages and hills in the background appear to be very far away (diminishing size); they look
smaller and distant from the rest of the main aspects of the painting. 5. Time and Motion– The
element of time is somewhat apparent in which the painting depicts events that happened during the
daytime because of the lighting and brightness used in the painting. The red and black colors
incorporated in the smoke and fire from the villages implement a sense of time, because although

Upon visiting the Reynolda House this weekend with a fellow classmate to work on our first outside–of–class writing assignment, this adapted outlook was especially apparent. The two of us wandered about the museum at a leisurely pace, and while I was already vaguely familiar with the layout from a previous visit as a little girl, three oil paintings from the Victorian era caught my eye. While they share an initial similarity with each other in their naturistic settings, they also differ in a myriad of ways. The first piece of art that I will be analyzing is The Old Hunting Grounds, an oil painting created by Worthington Whittredge in 1864. Whittredge largely limits his color pallete with mottled shades of green and brown typical of vegetation and forest life, though the canvas' focal point is dominated by a grove of illuminated birch trees.
